Özet
Bu çalışma ile Trakya Bölgesi içerisinde farklı lokalitelerden toplanan Sonchus L. cinsine (Asteraceae, Cichorieae) ait bitki örneklerinin morfolojik ve moleküler filogenetik araştırmaları yapılmıştır. Arazi çalışması sonucu toplanan Sonchus L. türlerine ait örneklerin (Sonchus asper (L.) Hill, Sonchus oleraceus L., Sonchus tenerrimus L.) morfolojik özellikleri stereo mikroskop ve taramalı elektron mikroskobu (SEM) kullanılarak ayrıntılı olarak incelenmiş olup, elde edilen sonuçlar fotoğraflarla desteklenerek kayıt altına alınmıştır. Moleküler çalışmalar için, örneklerden izole edilen gDNA'lar kullanılarak gerçekleştirilen, RAPD-PCR analizleri sonucunda türlerin ve lokalitelerin polimorfizm oranları belirlenmiş olup, en yüksek polimorfizm oranının S. asper türünde olduğu saptanmıştır. İzole edilen gDNA'ların ITS bölgelerine ait baz dizileri belirlenmiştir. Bu dizilere dayalı olarak oluşturulan filogenetik ağaçlar için mesafe temelli yöntem olarak UPGMA ve neighbor joining (NJ), karakter temelli yöntem olarak maksimum parsimoni (MP) metodları uygulanmıştır. Türkiye'de ilk defa S. asper, S. oleraceus, S. tenerrimus türlerinin morfolojik özellikleri, stereo mikroskop ve SEM kullanılarak ayrıntılı olarak incelenmiş olup, yine ilk defa RAPD-PCR analizleri ve ITS bölgesi dizi analizleri ile elde edilen moleküler sonuçlar ile karşılaştırılarak önemli veriler elde edilmiştir.
Morphological and molecular phylogenetic studies of plant samples belonging to the genus Sonchus L. (Asteraceae, Cichorieae) collected from different localities in Trakya Region were carried out with this study. The morphological characteristics of the samples belonging to Sonchus L. species (Sonchus asper (L.) Hill, Sonchus oleraceus L., Sonchus tenerrimus L.) collected as a result of the field study were investigated extensively using stereo microscope and scanning electron microscope (SEM) and the obtained results were supported with photographs and recorded. For molecular studies, polymorphism rates of species and localities were determined by RAPD-PCR analysis using gDNA isolated from samples, and the highest polymorphism rate was found in the S. asper. The base sequences of the ITS regions of the isolated gDNA were determined. For the phylogenetic trees based on these sequences, UPGMA and NJ were applied as distance-based method and MP methods as character-based method. Morphological characteristics of S. asper, S. oleraceus, S. tenerrimus species are elaborated in detail using a stereo microscope and SEM, RAPD-PCR analysis and molecular results obtained with the ITS region sequencing were compared with morphological results, for the first time in Turkey.
