Özet
Rho-kinaz yolağı nörogenez ve sinaptik plastisitede oldukça önemli bir role sahiptir. Bu çalışmada Rho-kinaz yolağının bağlamsal korku koşullanmanın edinim, konsolidasyon, geri çağırma, sönme ve rekonsolidasyon aşamalarındaki olası değişiminin ve Rho-kinaz yolağının inhibisyonun oluşturabileceği etkilerin incelenmesi amaçlandı. Toplam 303 adet Balb/c türü erkek fare kullanıldı. Farelere grup özelliklerine göre davranış deneylerinden 30 dk önce serum fizyolojik ya da 1, 3, ve 10 mg/kg dozda ROCK inhibitörü Y-27632, tek doz intraperitoneal yolla uygulandı. Daha sonra farelere rota-rod, bağlamsal korku koşullanma ve açık alan testleri yapıldı. Ayrıca bağlamsal korku koşullanma sürecinde ROCK enzim aktivitesinin nasıl değiştiğinin gösterilmesi amacıyla ROCK enzim aktivitesi ölçüldü. Bağlamsal korku koşullanmada geri çağırma ve sönme protokollerinde hipokampüste ROCK enzim aktivitesinin normal gruba göre arttığı görüldü. ROCK inhibitörü Y-27632 uygulanması, bağlamsal korku koşullanma deneylerinde davranışta herhangi bir etki oluşturmadı. Tüm gruplarda farelerin motor koordinasyonları açısından bir fark yoktu. Açık alan testinde ise edinim, konsolidasyon ve sönme protokollerinde özellikle 10 mg/kg dozda farelerin yürüme hızında ve buna bağlı olarak toplam gezilen mesafe verilerinde bir azalma olduğu görüldü. Ayrıca özellikle 10 mg/kg dozda Y-27632, bağlamsal korku koşullanmada tüm protokollerde ROCK enzim aktivitesini azalttı. Elde edilen bulgular bağlamsal korku koşullanmada geri çağırma ve sönme aşamalarında ROCK enzim aktivitesinin önemli olduğunu ancak bu yolağın inhibisyonunun korku belleğinin oluşumunu, geri çağırılmasını ya da sönmesini etkilemediğini göstermektedir.
The Rho-kinase pathway has a very important role in neurogenesis and synaptic plasticity. In this study, it was aimed to examine the possible changes of the Rho-kinase pathway in the acquisition, consolidation, recall, extinction and reconsolidation stages of contextual fear conditioning and to examine the effects of inhibition of the Rho-kinase pathway. A total of 303 Balb/c male mice were used. According to the group characteristics, 30 minutes before the behavioral experiments, saline or ROCK inhibitor Y-27632 at 1, 3, and 10 mg/kg doses was administered intraperitoneally as a single dose. Animals were then given rotarod to mice, contextual fear conditioning, and open field tests. In addition, ROCK enzyme activity was measured to show how ROCK enzyme activity changed during the contextual fear conditioning process. In contextual fear conditioning, ROCK enzyme activity in the hippocampus was increased compared to the normal group in recall and extinction protocols. Administration of the ROCK inhibitor Y-27632 had no effect on behavior in contextual fear conditioning experiments. There was no difference in motor coordination of mice in all groups. In the open field test, it was observed that there was a decrease in the walking speed of the mice, especially at the dose of 10 mg/kg, in the acquisition, consolidation and extinction protocols, and thus in the total distance traveled data. In addition, Y 27632 at a dose of 10 mg/kg decreased ROCK enzyme activity in all protocols in contextual fear conditioning. The finding show that ROCK enzyme activity is important in the recall and extinction stages in contextual fear conditioning, but inhibition of this pathway does not affect the formation, recall or extinction of fear memory.
