| dc.description.abstract | Bu tez çalışmasında Orchidaceaea familyasına ait Orchis laxiflora ve Orchis punctulata taksonlarının, sistematik ve moleküler filogenetik özellikleri sahip oldukları genomik DNA’larının 18S ve ITS1 bölgeleri sekanslanarak ortaya konulmuştur. Slika – jel yöntemiyle kurutulmuş bitki örneklerinin basal yaprak hücrelerinden genomik DNA’ları izole edilmiş ve 18S rDNA ve ITS1 bölgeleri önceden hazırlanmış primerler kullanılarak çoğaltılmıştır. Bu çoğaltılmış bölgelerin DNA sekanslaması yapılarak elde edilen veriler, bazı biyoinformatik ve filogeni programları ile çalışılarak bu bitkilerin moleküler düzeyde sınıflandırılmaları ilk kez ortaya koyulmuştur. CLUSTALW2 yazılımı kullanılarak üzerinde çalışılan bitkilerin diğer Orchidaceae üyeleri ile olan hizalamaları tespit edilirken MEGA 4,0 (STABLE) yazılımının Neighbour – Joining (Saitou ve Nei, 1987) metoduyla da filogenetik ağaçları oluşturulmuştur. Hizalama sonucu oluşan matrix – join tablosu ve filogenetik ağaç oluşturma işlemi sonucu elde edilen veriler klasik taksonomi verileri ile karşılaştırılmıştır. Bu tez çalışması ile O. laxiflora ve O. palustris’in A. coriophora, A. morio, A. champagneuxii ve A. pyramidalis ile ayrı bir grup oluşturduğu, O. punctulata’nın ise O. italica, O. mascula, O. militaris, ve O. purpurea ile ayrı bir grup oluşturduğu, ayrıca O. punctulata’nın Orchis cinsi içinde kalmaya devam etmesinin, O. laxiflora ve O. palustris’in ise Anacamptis cinsine aktarılmasının doğru olacağı sonuçlarına varılmıştır. | en_US |

| dc.description.abstract | In this study systematic and phylogenetic properties of the taxons of Orchis laxiflora and Orchis punctulata from the Orchidaceae family were determined by sequencing 18S rDNA and ITS2 regions of their genomic DNA. Genomic DNA’s were isolated from basal leaf cells of plant samples which were dessicated by silica-gel method. The 18S rDNA an ITS1 regions were amplified by previously prepared primers. The classification of these orchid species at the molecular level was accomplished for the first time, by applying bioinfomation and phylogenesis programs on the data obtained from the DNA sequences of these amplified regions. CLUSTALW2 program was used to determine the leveling of the species in question, with the other Orchidaceae species. Their phylogenetic trees were built by using the Neighbour-Joining (Saitou and Nei, 1987) method of MEGA 4,0 (STABLE) program. The data obtained from the matrix-join table and phylogenetic trees were compared with the classical taxonomical data. In conclusion, O. laxiflora and O. palustris were grouped together with A. coriophora, A. morio, A. champagneuxii and A. pyramidalis, in the same group, whereas O. punctulata was grouped with O. italica, O. mascula, O. militaris, and O. purpurea. Therefore it was proposed to keep O. punctulata to belong to the genera Orchis, but O. laxiflora and O. palustris to be transferred to the genus Anacamptis | en_US |
