Arpa’da kök büyümesi, antioksidatif enzim aktivitesi ve lipid peroksidasyonu üzerine arsenik’in etkisi

Özet
Bu çalışmada Arseniğin (As) Arpa bitkisinin (Hordeum vulgare) Sladoran ve Balkan 96 iki çeşidinde 10 µM (düşük ve non toksik doz) ve 50 µM (yüksek ve toksik doz) iki konsantrasyonunun çimlenme oranı, kök uzunluğu kuru ve taze ağırlık gibi fizyolojik parametreler değerlendirilmiş, arseniğin kök dokusunda Superoksit dismutaz (SOD), katalaz (CAT), askorbat peroksidaz (APX), Guaiacol peroxidase (GPX) gibi antioksidatif enzimlere ve oksidatif stresin önemli bir belirleyicisi olan lipit peroksidasyona etkileri incelenerek, arpa bitkisinin, arsenik toksisitesine biyokimyasal olarak cevabı araştırılmıştır.Arsenik, doz artışına paralel olarak arpada, her iki çeşitde kök büyümesini inhibe etmiş, köklerde taze ve kuru ağırlık belirgin şekilde azalmış, köklerde lipid peroksidasyonuna sebep olmuştur.Bunun yanı sıra, arseniğin oluşturduğu oksitatif stres karşısında arpa bitkisi kök dokusunda; süperoksit dismutaz (SOD), katalaz (CAT), askorbat peroksidaz (APX), guaiacol peroksidaz (GPX) gibi antioksidatif enzimlerin aktivitelerindeki değişiklerle cevap verilmiştir. APX ve CAT enzimlerinin aktivitelerinde anlamlı bir azalma (p<0.005) görülürken, SOD enzim aktivitesinde artma meydana gelmiştir. GPX enzim aktivitesinde ise anlamlı bir değişiklik meydana gelmemiştir.
Abstract
In this study, physiological parameters of Sladoran and Balkan 96 species of barley (Hordeum vulgare) Arsenic (As) such as germination rate, root length and dry and wet weight were evaluated in 10 µM (low and non-toxic dose) and 50 µM (high and toxic dose) two concentrations, its effect on antioxidative enzymes such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), Guaiacol peroxidase (GPX) in root tissue of arsenic and effect on lipid peroxidation which is an important determinant of oxidative stress were evaluated, the effect of barley on arsenic toxicity was analyzed in biochemical sense. Parallel to dose increase, arsenic inhibits root growth in both species of barley, dry and wet weight decreased distinctly, it caused lipid peroxidation in the roots.Besides, against oxidative stress caused by arsenic, barley reacted in root tissue with changes in antioxidative enzyme activities such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), Guaiacol peroxidase (GPX). While there was a significant (p<0.005) decrease in activities of APX and CAT enzyme activities, there was increase in SOD enzyme activities. There was no significant change in GPX enzyme activity.
