Özet
Bu çalışmada, bir noniyonik surfaktan(yüzey aktif madde) olan Triton X-100’ün farklı konsantrasyonlarının Allium cepa L.’nın kök uçlarında kök büyümesi, mitotik indeks, mitotik faz indeksi ve kromozom aberasyonları üzerindeki etkileri incelenmiştir. Kontrol uygulamaları musluk suyunda gerçekleştirilmiştir.Tüm denemelerde, 24 saat için musluk suyunda homojen köklenmiş soğanlar kullanılmıştır. Allium kök büyümesi inhibisyonu testinde, Triton X-100’ün etkili konsantrasyon (EC50) değeri yaklaşık % 0,125 g/L olarak bulunmuştur. Sitotoksisite testinde, soğanlar musluk suyu (kontrol) ve Triton X-100’ün EC50 değeri (% 0,125 g/L), EC50 değerinin yarısı (EC50/2=% 0,0625g/L) ve EC50 değerinin iki katı (EC50×2=% 0,25 g/L) konsantrasyonlarında 24, 48, 72 ve 96 saat süreyle bekletilmiştir. Triton X-100 uygulamalarında ortalama kök uzunluğu, kontrole göre önemli derecede azalmıştır (P<0,05). Bu uygulamalardan sonra, mikroskobik incelemeler için kök ucu örneklerinden preparatlar hazırlanmış, mitotik indeks, mitotik faz indeksi ve kromozom aberasyonları belirlenmiştir. Triton X-100 konsantrasyonunun artmasıyla mitotik indeks önemli düzeyde azaldığı, kromozom aberasyonlarının arttığı gözlenmiştir. Bu aberasyonlar, metafazda, tabla kayması (%22,13), yapışıklık (%12,02), vagrant kromozom (%5,62) ve C-mitoz (%3,29), anafazda, yanlış kutuplaşma (%18,96), vagrant kromozom (%14,7), köprü (%10,02), yapışıklık (%2,47) olarak belirlenmiştir. Abstract
In this study, the effects of nonionic surfactant Triton X-100 on root growth, mitotic index, mitotic phase index and chromosome aberrations on root tips of Allium cepa L. were investigated. The control treatments were performed in tap water. In all treatments, the onions which homogeneously rooted in tap water for 24 h were used. In Allium root growth inhibition test, effective concentration (EC50) value of Triton X-100 was found approximately % 0,125 g/L. In cytotoxicity test, onions were grown at tap water (control), and at EC50 (% 0,125 g/L), EC50 half of the (EC50/2=% 0,0625g/L) and double the EC50 (EC50×2=% 0,25 g/L) concentrations of Triton X-100 for 24, 48, 72 and 96 h. In concentrations of Triton X-100, mean of root growth reduced significantly compared to control (P<0,05). After these treatments, the slides were prepared from the root tip samples and mitotic index, mitotic phase index and chromosome aberrations were determined. It was observed that mitotic index was significantly decreased, chromosome aberrations induced by increasing the concentration of Triton X-100. These aberrations were detected such as plate shifting (%22,13), stickness (%12,02), vagrant chromosomes (%5,62) ve C-mitosis (%3,29) in metaphase, wrong polarization (%18,96), vagrant chromosomes (%14,7), bridge (%10,02), stickness (%2,47) in anaphase. Whereas the low rate of fragments in metaphase (%0,55) and fragments in anaphase (%1,09) were found.
