Bazı yabani ayçiçeği türlerinin çekirdek DNA içeriklerinin belirlenmesi
Özet
Ayçiçeği ülkemizin en önemli yağ bitkisidir. Yazlık ekilmesi ve ülkemizde
genelde kuru tarım yapılması nedeniyle, bazı yıllarda yaşanan kuraklık vb. abiyotik stres
koşullarının yanında orobanş, mildiyö vb. biyotik streslerden de fazlaca etkilenmektedir.
Bu nedenler ve son yıllarda artan küresel ısınma nedeniyle de, yeni geliştirilecek ayçiçeği
hibritlerinin bu stres koşullarına dayanıklı olması mutlak gereklidir. Bunun yanında,
hibrit ıslahında yüksek heterosis elde etmek için ebeveynlerin birbirine genetik
uzaklıklarını bilmek son derece yüksek önem arz etmektedir. Ayçiçeği (Helianthus spp)
cinsinin anavatanı Amerika olup, 37 çok yıllık ve 14 tek yıllık türden oluşmaktadır.
Yabani ayçiçeği (Helianthus spp) türleri biyotik ve abiyotik stres koşullarına dayanıklı
birçok gen kaynağına sahip olup, bu dayanıklılık genlerinin kültürü yapılan ayçiçeğine
aktarılmasında sürekli bir dayanıklılık sağlanması için büyük önem arz etmektedir. Bu
hedefler doğrultusunda çalışmanın amacı, ülkemiz koşullarında durumlarını belirlemek
için yurt dışından getirilen 13 türe ait tek yıllık 70 yabani ayçiçeği (Helianthus spp.) elde
edilmiş türünün çekirdek DNA içeriklerini flow sitometri ile belirleyerek, aksesyonların
ploidi düzeylerini ortaya çıkarmaktır. Çalışmada genetik materyal olarak özellikle, başta
orobanş olmak üzere, kuraklığa dayanıklılık ve ayçiçeğinde birçok hastalığa dayanıklı
genlere sahip tek yıllık yabani ayçiçeği türleri seçilmiştir. Bu genetik materyallerde, daha
sonra başta yüksek heterosis eldesi için ebeveynlerin seçimi olmak üzere, ayçiçeğinde
yabanilerle yapılacak diğer çalışmalara temel olması amacıyla, Flow Sitometri
yöntemiyle bu yabani türlerin DNA içerikleri ölçülerek ploidi düzeyleri belirlenmiştir.
Araştırma verilerine göre; çekirdek DNA içerikleri arasında değişimlerin ayçiçeği
türlerinin istatistiki olarak önemli olduğu saptanmıştır. Çalışmada elde edilen sonuçlara
göre, kullanılan ayçiçeği türlerinin ortalama 2C çekirdek DNA içerikleri 6,2 pg/2C (H.
neglectus ) ile 23,6 pg/2C (H. agrestis) arasında değişmektedir. Yapılan sitolojik
incelemelerde kromozomları sayılan iki türün de 2n=2X=34 ile diploid oldukları
görülmüştür. Çalışmada, doğru ve güvenilir bir yöntemle, dünyada ilk defa yabani
ayçiçeği türlerin DNA içerikleri belirlenmiştir. Sonuç olarak, uygun olan yöntem florasan
boya, standart ve örnek hazırlama tekniği kullanmak suretiyle, flow sitometri ile yapılmış
çekirdek DNA analizi sonucu elde edilmiş çekirdek DNA bilgisi cinsin içerisinde yer alan
türlerin taksonomik teşhisi, sınıflandırılması, ploidi analizi, genom yapı, ilişki ve
evrimlerinin incelenmesi, ıslah programları ve türler arası melezlerde hibritlerin
teşhisinde ve incelenmesinde yararlı olacağı saptanmıştır. Bunun yanında flow sitometri
analizinin ıslah ve taksonomik çalışmalarda morfolojik gözlemlerde tespit edilmesi
mümkün olmayan tür karışıklıklarının belirlenmesinde ve heterojen yapıda olanların
tespitinde kullanılabileceği ortaya konulmuştur. Sunflower is the most important oil crop in Turkey. Because of being spring crop and dry
cultivation, it is affected much by biotic stresses such as broomrape, mildew etc. as well
as abiotic stress conditions such as drought in some years. Therefore, for these reasons
and the increasing global warming recently, the new sunflower hybrids have to resistant
to these stress conditions. In addition, it is extremely important to know the genetic
distance of parents to obtain high heterosis in hybrid breeding. Sunflower genus
(Helianthus spp) is originated to America, consists of 37 perennials and 14 annual species.
Wild sunflower (Helianthus spp) species have many gene sources that are resistant to
biotic and abiotic stress conditions, and these resistance genes are of great importance for
providing continuous resistance to the cultivated sunflower. Based on these targets, the
aim of the study is to determine the ploidy levels of accessories utilizing flow cytometry
by measuring the core DNA contents of 70 wild sunflowers (Helianthus spp.) obtained
from 13 annual species brought from abroad. In this study, annual wild sunflower species
which have resistance to many diseases, broomrape and drought were selected as genetic
material. In order to be the basis of other studies with wild animals in sunflower,
especially the selection of parents for obtaining high heterosis in used genetic materials,
ploidi levels of these wild species were determined by Flow Cytometry method.
According to study data; the changes among the core DNA contents of sunflower species
were found to be statistically significant. Based on the results obtained in the study, the
average 2C core DNA contents of sunflower species ranged from 6.2 pg /2C (H.
neglectus) to 23.6 pg /2C (H. agrestis). Cytological examinations revealed that these two
species were diploid chromosomes with 2n = 2X = 34. The DNA contents of wild
sunflowers were determined for the first time in the world by an accurate and reliable
method in the study. Consequently, for taxonomic identification, classification, ploid
analysis, genome structure, relationship and evolution of the species within the genus,
nucleic DNA analysis obtained by flow cytometry using fluorescent dye, standard and
sample preparation techniques could be useful for breeding programs and also examining
and diagnosing of interspecific hybrids. Besides, it has been demonstrated that flow
cytometry analysis could be used in breeding and taxonomic studies to identify mixed
wild species that cannot be detected in morphological observations and to identify
heterogeneous ones among them.
