Öğe Determination of trace elements in soil and sunflower (Helianthus annuus L.) plant parts(Parlar Scientific Publications (P S P), 2007) Sabudak, Temine; Seren, Gulay; Kaykioglu, Gul; Dincer, Ali RizaTrace elements are currently of environmental concern. They are harmful to humans, animals and tend to bioaccumulate into the food chain. In this study, the concentrations of trace elements in soil and sunflower plant parts have been investigated. The concentrations of copper, manganese, iron, zinc in soil and sunflower plant samples collected from 5 different locations in Corlu, Turkey were determined by flame atomic absorption spectrometry after wet ashing. The maximum concentrations of trace elements per kg soil were 6.37 mg Cu, 0.53 10(3) mg Mn, 1.05 10(3) Mg Fe, and 36.07 mg Zn. The orders of their concentrations in soils and plant parts were found to be Fe>Mn>Zn>Cu. The maximum metal concentrations, except for Fe concentration, were observed in the leaves and seeds of plant parts. In addition, the sunflower leaves collected at locations 4 and 5 were observed for phytotoxic Mn concentration.Öğe The effect of melatonin on cadmium-induced renal injury in chronically exposed rats(Aves, 2009) Kaplan, Mustafa; Atakan, Irfan Huseyin; Aydogdu, Nurettin; Aktoz, Tevfik; Puyan, Fulya Oz; Seren, Gulay; Tokuc, BurcuObjective: This study was designed to investigate the effects of melatonin as an antioxidant, in prevention and treatment of cadmium (Cd)-induced renal toxicity in rats. Materials and methods: Fifty adult male Sprague-Dawley rats weighing 340 to 370 g were used. Renal toxicity was induced with the use of 200 mu g/ml of cadmium chloride in tap water. Ten rats were assigned to receive only tap water as the control group for three months and seven days. Two study groups were designed. To investigate the ability of melatonin to prevent Cd damage (Study 1), 10 rats received Cd and 10 rats received Cd plus 0.02% co-treatment melatonin in tap water for three months. To test whether melatonin would reverse CD-induced damage to the kidney (Study 2), 10 rats received Cd in tap water for three months+7 days, and 10 rats received Cd for three months followed by administration of high-dose (0.08%) melatonin in tap water for seven days. Results: Cadmium exposure significantly increased the kidney malondialdehyde (MDA) levels, as a marker of lipid peroxidation, decreased the kidney superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities, increased Cd concentrations in the renal cortex, but did not change glomerular filtration rate (GFR) and fractional excretion of sodium (FE-Na). In Study 1, melatonin decreased MDA levels, increased SOD, CAT and GSH-Px activities. In Study 2, melatonin decreased the kidney MDA levels, increased the SOD activity, but did not change the CAT and GSH-Px activities. Kidney accumulation of Cd did not change in both melatonin-treated groups. Histopathologically, Cd intake affected proximal tubules of the nephron more than the glomerular parts. Melatonin did not change these alterations. Conclusion: Our findings suggest that Cd-induced renal toxicity is related with oxidative stress and exogenously administrated melatonin might reduce the toxic effects of Cd on kidney without any reduction in tissue Cd burden.Öğe Effects of Arsenate and Arsenite on Germination and Some Physiological Attributes of Barley Hordeum vulgare L.(Springer, 2014) Sanal, Filiz; Seren, Gulay; Guner, UtkuArsenic (As) is toxic to plants and animals. We tested the effects of arsenite and arsenate (0-16 mg/L) on seed germination, and on relative root and shoot length, alpha-amylase activity, reducing sugars and soluble total protein contents, and malondialdehyde content in barley seedlings. We also measured As accumulation in barley stems and roots. The alpha-amylase activity, relative root and shoot length, and seed germination decreased with increasing concentrations of arsenate and arsenite. The reducing sugars content in barley seedlings increased after 4 days of growth on media containing As. In general, the protein content in roots and seedlings decreased with increasing doses of As. Arsenic in the tissues was quantified by hydride generation-atomic absorption spectrophotometry. To confirm the accuracy of the method, we analyzed the certified reference material WEPAL-IPE-168. The limit of detection was 1.2 mu g/L and the relative standard deviation was < 2.0 %.Öğe In vivo effects of curcumin and deferoxamine in experimental endometriosis(Wroclaw Medical Univ, 2017) Kizilay, Gulnur; Uz, Yesim Hulya; Seren, Gulay; Ulucam, Enis; Yilmaz, Ali; Cukur, Ziya; Kayisli, Umit AliBackground. Endometriosis is one of the most common chronic gynecological diseases. Objectives. The aim of the study was to examine the effects of curcumin and/or deferoxamine on cell proliferation in a rat model of endometriosis. Material and methods. Thirty female 12-week-old albino Wistar rats, weighing 200-250 g, were used in this study. All the rats underwent ovariectomy and 0.1-mg beta-estradiol 17-valerate pellets were placed intraperitoneally. An experimental model of endometriosis was created in all the animals. To create the experimental model, an approximately 1-cm long section of the uterus was taken, primarily from the right horn of the uterus. Autologous fragments were then placed between the peritoneum and muscle. The animals were divided into 3 groups: Group A, treated only with the vehicle used for curcumin and deferoxamine; group B, treated with curcumin (100 mg/kg body weight); and group C, treated with deferoxamine + curcumin (100 mg/kg body weight). After biopsy samples were obtained, the sections were stained with hematoxylin and eosin. Immunostaining for cytokeratin-7 and proliferating cell nuclear antigen (PCNA) was performed. Blood iron levels were measured using a Perkin Elmer AAnalyst 800 Atomic Absorption Spectrophotometer. Results. The endometrial implant size increased in Group A, but treatment with curcumin (p = 0.01) and deferoxamine + curcumin (p = 0.007) reduced the implant size. In ectopic endometrial epithelial cells, there were significant decreases in PCNA immunoreactivity between groups A and B (p = 0.044) and between groups A and C (p = 0.033). Conclusions. Treatment with curcumin alone and/or in combination with deferoxamine contributed to a reduction in implant size and cell proliferation in a rat endometriosis model. Iron-chelating agents may act in the same manner when used in women with endometriosis; however, further studies from different perspectives are still needed.Öğe Serum, urinary and stone zinc, iron, magnesium and copper levels in idiopathic calcium oxalate stone patients(Springer, 2007) Atakan, Irfan H.; Kaplan, Mustafa; Seren, Gulay; Aktoz, Tevfik; Gul, Hatice; Inci, OsmanMany theories have been put forward to explain the mechanism of stone formation and growth. The aim of this study was to investigate the urinary, serum and stone levels of zinc, iron, magnesium, and copper in patients with calcium oxalate stones and to investigate urinary and serum element levels in healthy controls and to find a possible connection between the elements and calcium oxalate stone formation. A total of 104 patients with calcium oxalate stones ranging in age from 3 to 79 years (mean 44.0 +/- 18.1) and 77 healthy controls ranging in age from 18 to 77 (mean 44.2 +/- 17.9) were included in this study. The mean urinary iron and copper levels in stone patients were significantly higher than healthy controls (P = 0.000). The mean urinary zinc and magnesium levels in healthy controls were significantly higher than stone patients (P = 0.000). There was no significant difference in the serum levels of magnesium and copper in stone patients and healthy controls. Serum zinc and iron level were significantly high in healthy controls as compared to stone patients. Each stone had all 4 elements. Zn and Mg have inhibitory effect on calcium oxalate stone formation. Fe and Cu could be promotor of the calcium oxalate stone formation.
