Öğe Airborne Alternaria and Cladosporium species and relationship with meteorological conditions in Eskisehir City, Turkey(Springer, 2008) Erkara, Ismuhan Potoglu; Asan, Ahmet; Yilmaz, Veysel; Pehlivan, Sevil; Okten, Suzan SaricaAlternaria and Cladosporium, known as the most allergenic spores were first collected by means of Durham gravimetric sampler from Eskisehir atmosphere from January 1, 2000 to December 31, 2001. The daily, monthly and annual variations in spores/cm(2) of Cladosporium and Alternaria were recorded. During this period, a total of 10.231 spores belonging to Cladosporium and Alternaria genera were recorded. Of these spores, 5,103 were identified in 2000 and 5,128 in 2001. While 63.09% of the total spores were those of Cladosporium, 36.91% were of Alternaria. Relationships between airborne fungal spore presence and meteorological conditions were statistically investigated. A Shapiro-Wilk test revealed that the airborne Cladosporium and Alternaria spores differed from a normal distribution. Thus, a Friedmann test was performed followed by a Pearson Correlation Analysis. The effects of rainfall, temperature and wind speed on Cladosporium and Alternaria numbers were non-significant according to the sites and months (p>0.05), but the effects of relative humidity on Cladosporium and Alternaria numbers were significant (p<0.01). Spore concentrations reached to their highest levels in May 2001.Öğe Airborne and soilborne microfungi in the vicinity Hamitabat Thermic Power Plant in Kirklareli City (Turkey), their seasonal distributions and relations with climatological factors(Springer, 2010) Asan, Ahmet; Okten, Suzan Sarica; Sen, BurhanSoil and air samples of seven different localities around Hamitabat Thermic Power Plant, 10 km far away from Luleburgaz/Kirklareli (Turkey), were taken between the years 2003 and 2004 with seasonal intervals. The samples were brought to the laboratory and their microfungal identifications were done. From the air samples, 737 microfungi colonies were isolated comprising 26 species belonging to eight genera. From soil samples, 170.6 x 10(4) colony-forming unit (CFU)/g was isolated from 33 species belonging to 16 genera. The most isolated genus from air samples was Alternaria (324 CFU, 43.96%), followed by Cladosporium (208 CFU, 25.52%) and Phoma (44 CFU, 5.40%). Penicillium was the most isolated genus from the soil samples with a value of 560,000 CFU/g (32.8%), followed by Fusarium (226,000 CFU/g, 13.12%) and Aspergillus (154,000 CFU/g, 9.03%). Among these species, Alternaria citri and Alternaria alternata are the most abundant species in air with 164 and 107 CFU, respectively, whereas Fusarium graminearum and Penicillium citrinum are the most abundant species in soil with CFU per gram values of 17.8 x 10(4) and 1.3 x 10(5). Correlation analysis was applied to determine whether or not there was a relationship between colony number of isolated fungal genera and meteorological factors. Some parameters of soil samples' incontent during the research period were calculated using a computer analysis program. From the air samples, a positive correlation was found between relative humidity and Alternaria colonial counts and Cladosporium spore counts (r = 0.912 and r = 1.000, respectively). Similarly, with the analysis of soil samples, a positive correlation between colonial counts of Alternaria and soil pH and a positive correlation between colonial counts of Aspergillus and Penicillium and salt percentage concentration of soil were found.Öğe AIRBORNE FUNGAL BIODIVERSITY IN INDOOR AND OUTDOOR AIR OF THREE MOSQUES IN EDIRNE CITY, TURKEY(Parlar Scientific Publications (P S P), 2018) Tikvesli, Melek; Asan, Ahmet; Gurcan, Saban; Sen, BurhanThe purpose of our study is to determine the fungal numbers and type of air fungi in indoor and outdoor air ambient of three different mosques in Edirne City. Research materials were taken from indoor and outdoor environments of three different mosques in Edirne once a month during 12 months between October 2008 and September 2009. Samples were taken (in indoor and outdoor environments, 1 minute each of them and 100 liters) from selected stations by using Air Sampling Device (Merck Millipore Air Sampler, Germany). Fungi samples were taken from indoor and outdoor environments of each station approximately 1 m above. Identification of fungi carried out by classical methods such as morphology and colonial characteristics of fungi. 22 fungal genera and 69 fungal species belonging these genera were determined. Cladosporium genus was the most abundant among airborne fungi with 2338 CFU/m(3) (52.90%) It was followed by Penicillium genus with 875 CFU/m(3) (16.55%), Alternaria genus with 457 CFU/m(3) (8.64%) Aspergillus genus with 216 CFU/m(3) (4.08%). About Fungal genera and total microfungi concentrations during the study period, maximum value of fungal concentrations in indoor air was determined as 609 CFU/m(3) (11.52%) in mosque C in September and maximum value in outdoor air was determined as 961 CFU/m(3) (18,18%) at the same place in September. It was observed that fungi concentrations in three mosques were in healthy limit values. Considering whether there is a relationship between fungi and meteorological factors, it was determined that the Alternaria genus was not affected by any meteorological conditions.Öğe Airborne Fungal Concentrations in East Patch of Edirne City (Turkey) in Autumn Using Two Sampling Methods(Trakya Üniversitesi, 2005) Okten, Suzan Sarıca; Asan, Ahmet; Tungan, Yasemin; Ture, MevlutGece ve gündüz saatlerinde fungal konsantrasyonlar değişiklik gösterebilir. Kuru sporlar havaya gündüz salınırken nem oranı daha yüksek olan sporlar gece salınmaktadırlar. Türkiye’de havayla taşınan funguslar ile ilgili çalışmalar çok azdır ve yalnızca bazı şehirlerde günlük ve haftalık incelemeler yapılmıştır. Bu çalışmada Edirne şehri günlük fungal konsantrasyonlarının belirlenmesi amaçlanmıştır. Örnekler yerden 11 m yükseklikten alınmıştır. Örneklemeler 11 Ekim 2002 – 15 Kasım 2002 tarihleri arasında, hergün aynı saatlerde olmak üzere birer haftalık zaman dilimlerinde sabah 08.00 ve akşam 20.00’de yapılmıştır. Fungus izolasyonu için yerçekimine dayalı petri plak metodu ve volumetrik örnekleme metodu kullanılmıştır. Sıcaklık ve nısbi nem iklimsel faktörlerin fungal sporları etkileyip etkilemedikleri istatistiki olarak incelenmiş ve fungus sayıları, nısbi nem ve sıcaklık arasında korelasyon bulunmuştur. 42 petri plağına izole edilen toplam 800 fungus kolonisi bulunma sıklıkları belirlenerek cins düzeyinde tanımlamaları yapılmıştır. Volumetrik metot ile m3 de 9337 cfu mikrofungus belirlenmiştir. En baskınları Alternaria, Penicillium ve Trichoderma olan 7 fungus cinsi teşhis edilebilmiştir.Öğe Airborne Fungal Concentrations of Morning and Evening in East Patch of Edirne City Using Two Sampling Methods(Trakya Üniversitesi, 2007) Ökten, Suzan Sarica; Asan, Ahmet; Sabuncuoğlu, Yıldız; Yavuz, EbruEylül- Kasım 2002 tarihleri arasında sabah ve akşam saatlerinde havayla taşınan fungusların konsantrasyonlarının değişimlerini belirlemek amacı ile ayda üç kez olmak üzere fungal örnekleme yapılmıştır. Bu amaç için iki metot uygulanmıştır. Örnekler, Yerçekimine Dayalı Petri Plak Metodu ile, Rose Bengal Streptomisin agar içeren petri plakları 15 dakika havayla temas ettirilerek ve Volumetrik bir örnekleyici kullanarak alınmıştır. Alternaria, Aspergillus, Cladosporium, Penicillium, Rhizopus, Scopuloriopsis, Trichoderma, Ulocladium cislerine ve Dematiaceae familyasına ait 9680 koloni gözlenmiştir. Bunlardan Cladosporium, Alternaria ve Penicillium cisleri Yerçekimine Dayalı Petri Plak Metodu için 120, 149 ve 4, Volumetrik örnekleme için de 3133, 2733 ve 1134 koloni sayıları ile baskın cinsleri oluşturmuşlardır. Değişkenlerin normal dağılıma uygun olup olmadıklarını belirlemek için Kolmogorow-Simirnov testi uygulanmıştır. Daha sonra yapılan Korelasyon analizi sonucunda Volumetrik metotla izole edilen cinslerin sabah ve akşamki koloni sayıları arasında anlamlı bir ilişki bulunmuştur (r= 0.489 ). Yerçekimine Dayalı Petri Plak Metoduna göre izole edilen cinslerin sabah ve akşamki koloni sayıları arasında da yine anlamlı bir ilişki bulunmuştur (r= 0.667). Her iki metoda göre izole edilen cinslerin sabah ve akşamki koloni sayıları istatistiki olarak analiz edilmiş ve aralarında bir ilişki olduğu saptanmıştır (r= 0.486 ve r= 0.761).Öğe AIRBORNE FUNGAL DIVERSITY INSIDE A NURSING HOME IN EDIRNE, TURKEY(Parlar Scientific Publications (P S P), 2017) Yilmaz, Oznur; Asan, Ahmet; Aydogdu, Halide; Sen, BurhanWe aimed to determine the indoor airborne fungal concentration and diversity of a nursing home in Edirne City (Turkey). We sampled indoor airborne fungal sampling two times a month during 6 months period from 9 different places of nursing home. Also we investigated distribution of fungi as monthly seasonal and relationship with meteorological factors. We determined 9.520 CFU/m(3) fungal colonies by aspiration 21.600 L of air using 216 Petri plates for fungal isolation. The highest fungal counts were found in dining hall and the lowest in lounge. Also the highest fungal counts found in April and lowest in December. We determined 80 fungal species belonging to the 31 genera. The most appearing fungal genera were Penicillium, Cladosporium and Aspergillus and Penicillium genus had highest species diversity with 34 species. We determined Penicillium, Cladosporiurn, Aspergillus, Mycelia sterilia and Alternaria in all months. Penicillium and Aspergillus genera were the most appeared in April and Cladosporium was in May. Correlation of Alternaria, Aspergillus and Cladosporium genera with temperature was found significant as statistics (*p<0.05). There are living old humans whose have generally health problems in nursing homes and determination of indoor airborne fungi in these places are important for prevent allergic reactions and potential fungal infections.Öğe Airborne fungi and bacteria in indoor and outdoor environment of the Pediatric Unit of Edirne Government Hospital(Springer, 2012) Okten, Suzan; Asan, AhmetThis study was performed between January 2004 and December 2004 in 13 stations in the Pediatric Unit of Edirne Government Hospital in order to determine the outdoor and indoor airborne microfungal and bacterial contents. The results of air samplings revealed that 1,376 microfungal and 2,429 bacterial colonies in total were isolated. The isolated microfungal specimens were identified and 65 species from 16 genera were determined. Among these, the most frequent genus was Cladosporium with 462 colonies (33.58%) followed by Alternaria with 310 (22.53%) and Penicillium with 280 (20.35%) colonies. The isolated bacterial samples were grouped based on their Gram-staining properties. The most frequent ones were Gram (+) cocci with 1,527 colonies (62.87%) followed by Gram (+) bacilli with 828 colonies (34.09%) and Gram (-) bacilli with 74 colonies (3.05%). Staphylococcus, Bacillus, Corynebacterium, and Microccus appeared to be the common genera isolated for all months. Statistical analyses were performed in order to see if there existed a relationship between meteorological conditions and the microfungal and bacterial species and their concentrations.Öğe Airborne fungi in child day care centers in Edirne City, Turkey(Springer, 2008) Aydogdu, Halide; Asan, AhmetThe purpose of this study was to determine the concentration, in terms of monthly and seasonal distribution and in relation to meteorological factors, of indoor and outdoor microfungi at selected sites in several child day care centers in the city of Edirne, Turkey. Samples were collected at one month intervals over a period of 12 months between January-December 2004, by exposing petri plates containing Peptone Dextrose Agar with Rose-Bengal and Streptomycin medium to the air for 10-15 min. A total of 2,071 microfungal colonies were counted on 192 petri plates. Thirty microfungal genera (Acremonium, Alternaria, Arthrinium, Aspergillus, Bahusakala, Beauveria, Ceuthospora, Chaetomium, Cladosporium, Curvularia, Drechslera, Epicoccum, Eurotium, Fusarium, Mycotypha, Myrotechium, Paecilomyces, Penicillium, Pestalotiopsis, Phoma, Ramichloridium, Rhizopus, Scopulariopsis, Stachybotrys, Stemphylium, Torula, Trichoderma, Trichothecium, Ulocladium, Verticillium) and 75 microfungal species were isolated from the air indoor and outdoor of the day care centers. The dominant microfungal genera were Cladosporium, Penicillium and Alternaria (44.11%, 18.94%, 14.67% of the total respectively), while the genus with the most species richness was Penicillium (26 species). Alternaria, Cladosporium, Penicillium and non-sporulating microfungi were found every month. Cladosporium was the dominant genus in both indoor and outdoor air. Although the predominant genus was the same in both indoor and outdoor air, Cladosporium was followed by Penicillium, Alternaria and Aspergillus genera in indoor air and by Alternaria, Penicillium and Aspergillus genera in outdoor air. While a positive correlation was found between the concentration of monthly outdoor microfungi and monthly average temperature, a negative correlation was found between the concentration of monthly outdoor microfungi and monthly average wind velocity. Also, some relationships were found between the monthly concentrations of the most predominant microfungal genera (Cladosporium, Penicillium and Alternaria) and various meteorological factors.Öğe Airborne microfungi in Oncology Service of Medical School Hospital of Trakya University(Sage Publications Ltd, 2015) Okten, Suzan; Sen, Burhan; Asan, Ahmet; Bahadir, NurcanThis study was performed in six different stations - drug preparation, chemotherapy, radiotherapy and three different patient rooms - in Oncology Service of Trakya University Medical Hospital in Edirne. Indoor air samples of equal volumes of 100 L were taken twice a month during the six-month study period. The analysis of samples showed that 8540 microfungi colonies were present in a total of 7.2 m(3) air sample. November and April were found to be the months during which the highest numbers of fungal spores were isolated with 2570 CFU/m(3) and 2060 CFU/m(3), respectively, while the lowest spore counts were isolated during January (730 CFU/m(3)). Penicillium was found to be the most commonly isolated fungal genus with 2950 CFU/m(3)/spore followed by Cladosporium with 2110 CFU/m(3). The other fungal genera isolated during the study are Aspergillus (640 CFU/m(3)), Alternaria (200 CFU/m(3)), Trichoderma (70 CFU/m(3)) and Rhizopus (40 CFU/m(3)). Among the most frequently isolated fungal species, Penicillium brevicompactum was the most common species with 960 CFU/m(3) followed by Cladosporium cladosporioides with 930 CFU/m(3). Aspergillus fumigatus, the fungal species of paramount importance in terms of human health, was also reported with a 410 CFU/m(3). Statistical analyses were performed in order to reveal the relationship between microfungal colony numbers isolated and station temperature and relative humidity.Öğe The antimicrobial effect of various formulations obtained from Fomes fomentarius against hospital isolates(2019) Gedık, Gulsah; Dulger, Gorkem; Asan, Hulya; Özyurt, Anıl; Allı, Hakan; Asan, AhmetThe purpose of this study was to investigate the antimicrobial activity of formulations of F. fomentarius. The antimicrobial efficacy levels of extracts of F. fomentarius prepared from ethanol, water and of various formulations (emulsion, ointment, paste, cream and gel) were determined. For this purpose, the disk diffusion method was used to test the extracts for antimicrobial activity against hospital isolates including Klebsiella pneumoniae, Acinetobacter baumannii, Staphylococcus aureus, vancomycin-resistant enterococci (VRE+), Escherichia coli, Candida krusei, C. albicans, C. tropicalis, C. guilliermondii and C. glabrata. Some standard antibiotics were used for comparison. The ointment formulation (No. 4) showed a good antimicrobial effect on bacteria compared to the antibiotics (9.6 - 16.1 mm.). The extracts were found to be more effective than the antibiotics against Candida species. Formulation No. 4 was considered to be more effective because of the protective nature of its cetyl alcohol and sodium lauryl sulfate (SLS) content . F. fomentarius ethanol extract, F. fomentarius water extract and gel formulation showed similar activity; however, other formulations (emulsion, paste, cream) were generally more effective due to the stabilizing effects of the polymers, oil and alcohols present in their preparations. The findings of this study establish the possibility of discovering new clinically effective antibiotic drugs and could be useful in understanding the relationship between traditional remedies and modern medicines.Öğe Bazı Kullanılmış Kozmetik Ürünlerinde Fungal Çeşitlilik(2021) Elbasan, Döndü Gümüşkaya; Asan, Ahmet; Ökten, SuzanBu çalışmada, çeşitli saklama koşullarında kullanılmış kozmetik ürünlerde bulunan mikrofungusların morfolojik yöntemlerle cins düzeyinde tanımlamaları yapılarak, ürünlerdeki mikrofungal çeşitliliğin tespiti amaçlanmıştır. Çalışmada; farklı süre ve koşullarda kullanılmış kozmetik ürünler kullanıcılardan temin edilmiştir. Bu ürünlerin her birinden 5 er adet olmak üzere ruj, allık, rimel, göz farı, göz kalemi, pudra, göz makyaj temizleme ürünü ve roll-on olarak gruplandırılmış ve toplam 40 adet ürün incelenmiştir. Mikrofungus izolatları saf kültür olarak elde edilip yatık PDA besiyerine pasaj alınarak 25°C'de 7 gün inkübe edilmiş ve daha sonra stok kültür olarak +4°C'de saklanmıştır. İnkübasyon sonucunda izole edilen mikrofungusların makroskobik ve mikroskobik incelemeleri yapılarak cins düzeyinde tespiti yapılmıştır. Çalışma sonucunda baskın fungus % 57.9 oranında Penicillium (Penisilyum) cinsi, % 23.5 oranında Aspergillus (Asper), % 10.3 oranında Cladosporium (Havaküfü) ve % 8.3 oranında Alternaria (Arıküfü) cinsleri tespit edilmiştir. Çalışmada incelenen kozmetik ürünlerden en fazla fungal üreme görülen ürün grupları sırasıyla 76 koloni ile far, 50 koloni ile ruj ve 49 koloni ile pudradır.Öğe Biodiversity of Penicillium species isolated from Edirne Söğütlük Forest soil (Turkey)(2019) Kolanlarlı, Tugba Korur; Asan, Ahmet; Sen, Burhan; Ökten, SuzanWe studied Penicillium species isolated from forest soil and aimed to be detected with morphologically, colonial-molecular methods and species diversity. Soil sample was taken Edirne Söğütlük Forest in October, 2015 and 10 cm deep under aseptic conditions and Waksman’s ‘Soil Dilution Method’ was used for the isolation of Penicillium species; some books and articles have been used for morphological and colonial identification. The service was purchased for the molecular diagnosis of fungi and the company used the Bio-Speedy ™ Fungal DNA Isolation Kit for isolation. As PCR Kit, Bio-Speedy™, yeast and mould real-time PCR rapid detection kit was used. The obtained sequences were subjected to BLAST analysis similar sequences using the gene bank and to draw phylogenetic tree, used Mega 7 software. Twelve species belonging to the genus Penicillium were identified; these are: P. chrysogenum, P. citrinum, P. commune, P. manginii (current name: Penicillium atrosanguineum) , P. ubiquetum (New record for Turkey), P. camemberti, P. sanguifluum, P. janczewskii, P. canescens, P. roseopurpureum, P. steckii (current name: Penicillium citrinum), P. hordei.Öğe Checklist of Cladosporium Species Reported from Turkey(2016) Asan, Ahmet; Özkale, Evrim; Kalyoncu, FatihCladosporium species are common in nature (air, soil, etc.) and can be allergenic on human. The purpose of this study is to document the Cladosporium species isolated from Turkey with their subtrates and/or habitat. This checklist reviews approximately 114 published findings and presents a list of Cladosporium species. According to the present publications, 33 Cladosporium species have been recorded with various subtrates/habitats in Turkey. Among these species, Cladosporium herbarum and Cladosporium cladosporioides are the most common species reported from Turkey. The other species are Cladosporium acaciicola, C. aecidiicola, C. apicale, C. asterinae, C. brassicae, C. britannicum, C. carpophilum, C. chlorocephalum, C. colocasiae, C. cucumerinum, C. elatum, C. fulvum, C. gallicola, C. macrocarpum, C. musae, C. nigrellum, C. obtectum, C. orchidaceraum, C. orchidis, C. ornithogali, C. oxysporum, C. ramotenellum, C. resinae, C. sinuosum, C. sphaerospermum, C. spongiosum, C. tenuissimum, C. trichoides, C. uredinicola and C. variabile. The oldest literature is belonging to the year of 1952Öğe Comparison of Quartile Scores of Mycology Journals Covered by Web of Science and SCImago SCOPUS Databases(2021) Asan, Ahmet; Giray, GülayMillions of papers are published in scientific journals every year. For example, 3,194,821 papers were published only in the Clarivate Analytics Web of Science Core Collection Database in 2020. However, the citation rates, h index and Q scores of journals are different from each other. While important scientific results are published in some journals, which are the basis of the literature, other journals may be weak and their citation rates may low. The data used in this study are taken from Clarivate Analytics Web of Science and SCImago SCOPUS databases. In this study, we tried to take attention some data about Q scores of scientific journals. The Q scores of scientific journals have become important in academic appointments in Turkey and also become one of the quality indicators of journals. For this purpose, some data of 30 mycology journals were taken from Clarivate Analytics Web of Science and SCImago SCOPUS databases. Q scores, h index, impact factor scores and some other data of these mycology journals were given in table and analyzed. While there are 8 journals in the Q1 category covered by Web of Science, there are 20 journals in the Q1 category covered SCImago SCOPUS. This is due to the different number of journals and subject categories in both databases. Among 30 mycology journals, Fungal Diversity was the journal with the highest impact factor and Q1 category with 15.39 in the year of 2019. The journal with the highest h index score is Mycologia with a score of 128. In addition, number of papers with Turkey addressed published in 30 mycology journals are given (790 publications, 638 full article) in this study.Öğe Determination of Aflatoxin M1 on Follow-on Milk by ELISA Method(2017) Tuz, Meltem Kaya; Ökten, Suzan; Asan, AhmetÖnemli bir besin kaynağı olan süt, bebek beslenmesinde kullanılmakta ve tüm dünyada tüketilmektedir. Devam sütlerine geçtiği bilinen kimyasal kontaminantlardan biri mikotoksinlerdir. Küfler tarafından üretilen ve devam sütlerine geçebilen mikotoksinler bebekte sağlık riski oluşturabilir. Mikotoksinlerin toksik etkilerinin önlenmesi için bebeğin normal büyüme ve gelişimi için en önemli besinlerden biri olan devam sütündeki aflatoksin M1 düzeylerinin belirlenmesi önemlidir. Süt kontaminantı olarak aflatoksinlerin kontrolü ile ilgili sosyal koruma projelerinin ve devam sütleri ile ilgili farkındalığın arttırılması halk sağlığı korunması açısından önem arz eder. Çalışmamızda, yeni doğanların diyetlerinde aldığı ürün olan devam sütlerinde, aflatoksin M1'in varlığı ve halk sağlığı bakımından önemi araştırılmıştır.Bu amaçla, 7 farklı firmadan, farklı seri numaralarına sahip süt örneklerini içeren 60 adet devam sütü toplanmıştır. Çalışmamızda Enzyme-linked immunosorbent assay (ELISA) yöntemi kullanılarak devam sütlerinde aflatoksin M1miktarları araştırılmıştır. Avrupa Birliğine uyum çerçevesinde hazırlanmış olan 2008/26 sayılı gıda maddelerindeki kontaminasyonların maksimum limitleri hakkındaki Türk Gıda Kodeksi (TGK) Tebliğinde; çiğ süt, UHT süt ve süt içeren ürünlerin yapımında kullanılan sütlerde maksimum AFM1 miktarının 0.05µg/kg (parts per billion) olması gerektiği belirtilmiştir. Aflatoksin M1 konsantrasyonları hesaplandığında; <0.05 parts per billion konsantrasyon aralığında Aflatoksin M1 içeren 37 örnek, >0.05 ppb konsantrasyon aralığında Aflatoksin M1 içeren 23 örnek tespit edilmiştir. Sonuç olarak Türk Gıda Kodeksi limit değerini aşan 23 örnek halk sağlığı açısından önem arz etmektedir. Beslenmede devam sütünün sıklıkta tüketildiği ve önemli bir rol oynadığı bebek yaş grubunda, bu ürünler piyasaya çıkmadan önce Aflatoksin M1 varlığının saptandığı daha kapsamlı araştırmaların yapılmasının uygun olduğu düşünülmektedirÖğe Devam sütlerinde aflatoksin M1 varlığının ELISA yöntemiyle tespit edilmesi(Trakya Üniversitesi Fen Bilimleri Enstitüsü, 2017) Kaya Tuz, Meltem; Asan, Ahmet; Ökten, SuzanÖnemli bir besin kaynağı olan süt, bebek beslenmesinde kullanılmakta ve tüm dünyada tüketilmektedir. Devam sütlerine geçtiği bilinen kimyasal kontaminantlardan biri mikotoksinlerdir. Küfler tarafından üretilen ve devam sütlerine geçebilen mikotoksinler bebekte sağlık riski oluşturabilir. Mikotoksinlerin toksik etkilerinin önlenmesi için bebeğin normal büyüme ve gelişimi için en önemli besinlerden biri olan devam sütündeki aflatoksin M1 düzeylerinin belirlenmesi önemlidir. Süt kontaminantı olarak aflatoksinlerin kontrolü ile ilgili sosyal koruma projelerinin ve devam sütleri ile ilgili farkındalığın arttırılması halk sağlığı korunması açısından önem arz eder. Çalışmamızda, yeni doğanların diyetlerinde aldığı ürün olan devam sütlerinde, aflatoksin M1’in varlığı ve halk sağlığı bakımından önemi araştırılmıştır. Bu amaçla, 7 farklı firmadan, farklı seri numaralarına sahip süt örneklerini içeren 60 adet devam sütü toplanmıştır. Çalışmamızda Enzyme-linked immunosorbent assay (ELISA) yöntemi kullanılarak devam sütlerinde aflatoksin M1miktarları araştırılmıştır. Avrupa Birliğine uyum çerçevesinde hazırlanmış olan 2008/26 sayılı gıda maddelerindeki kontaminasyonların maksimum limitleri hakkındaki Türk Gıda Kodeksi (TGK) Tebliğinde; çiğ süt, UHT süt ve süt içeren ürünlerin yapımında kullanılan sütlerde maksimum AFM1 miktarının 0.05µg/kg (parts per billion) olması gerektiği belirtilmiştir. Aflatoksin M1 konsantrasyonları hesaplandığında; <0.05 parts per billion konsantrasyon aralığında Aflatoksin M1 içeren 37 örnek, >0.05 ppb konsantrasyon aralığında Aflatoksin M1 içeren 23 örnek tespit edilmiştir. Sonuç olarak Türk Gıda Kodeksi limit değerini aşan 23 örnek halk sağlığı açısından önem arz etmektedir. Araştırma, şubat 1986 ile şubat 1987 tarihleri arasında yürütülmüştür. Bu amaçla Edirne îli ve bu ile bağlı köylerden getirtilen 24 örnek incelenmiştir. örnekler, "Su Ağarı" metoduyla incelenmiştir. Böylelikle yüksek oranda fungus çıkışına imkan sağlanmış ve çıkan fungusların incelenme ve izolasyonları daha kolay olmuştur. Ayrıca Pusarium türlerine ait tüm oluşumları elde etmek amacıyla, Prinç Lapası, Yonca Sapı, Havuç Agar, Yulaf Unu Agar, Bira Şırası Agar ve Pata tes Sukroz Agar "besi ortamları kullanılmıştır. Değişik besi ortamlarına aşılanan Fusarium türleri, yapı sal olarak ayrıntılı biçimde İncelenmiştir.. Ayrıca yukarıda sözü edilen besiyerlerine aşılanmış Fuaarium türlerinin renk değişimle ri, ortalama 10 gün aralıklarla not edilmiştir. Araştırma sonucunda, Alternaria, Aspergillus, Cladosporium, Helminthosporium, Penicillium, Rhizopus ve Fnsarium cinsleri ile, P. equiseti, F. graminearum ve F. moniliforme türleri saptanmıştır. Bu 3 tür, mısırda tohumla taşınan funguslar arasında, Türkiye* de ilk defa bulunmuştur. Bu 3 tür içinde, F. moniliforme en yüksek oranda (% 71.86) bulunmuştur.Öğe Edirne İli Söğütlük Ormanı Toprağından İzole Edilen Aspergillus Türlerinin Biyoçeşitliliği(2018) Ayan, Eda Gizem; Asan, Ahmet; Sen, Burhan; Okten, SuzanÇalışmamızda, Edirne ili orman topraklarından izole edilen Aspergillus türlerinin morfolojik-koloniyal ve moleküler yöntemlerle teşhis edilmesi amaçlanmıştır. Bu amaçla, Edirne Söğütlük Ormanından, 2016 yılı Mart ayında Brown’un metodu kullanılarak toprak örnekleri alınmıştır. Topraktan fungus izolasyonu için “Toprağı Sulandırma Metodu” kullanılmıştır. Morfolojik çalışmalar için CZ, CYA, CY20S, MEA besiyerlerine yapılan ekimlerden sonra, mikroskobik ve makroskobik karakterler incelenmiştir. Moleküler çalışmalar için ise sırasıyla DNA izolasyonu, calmodulin gen bölgesini hedefleyen PCR işlemleri, PCR ürününün saflaştırılması, dizi analizi ve filogenetik analiz aşamaları uygulanmıştır. Çalışmalar sonucunda 7 adet Aspergillus türü tespit edilmiştir. Bu türler, Aspergillus affinis (Türkiye için yeni kayıt), Aspergillus awamori, Aspergillus carbonarius, Aspergillus dimorphicus , Aspergillus europaeus (Türkiye için yeni kayıt), Aspergillus spelaeus (Türkiye için yeni kayıt) ve Aspergillus fischeri’dir.Öğe Edirne ili topraklarından izole edilen Aspergillus Mich.ex Fr. ve penicillium Link ex Fr. türleri üzerinde taksonomik ve ekolojik araştırmalar(Trakya Üniversitesi, 1992) Asan, Ahmet; Ekmekçi, SanverBu çalışmada, Edirne İl Sınırları içinde kalan ve habitatları farklı olan toplam 7 istasyon seçilmiş ve bu istasyonlarda» 4 mevsim boyunca 10 cm. derinlikten toprak örnekleri alınarak, Asper gillus Mich. &x Pr. ve Penioillium linkex Pr. florası araştırılmıştır. "Toprağı Sulandırma Metodu" kullanılarak sonuçlandırılan araştırmada, her iki cinse ait 23 tür ve 2 varyete tesbit edilmiştir. Bunlardan 7 tür ve 2 varyete Aspergillus Mich, ex Pr., 16 tür ise Penioillium Link ex Fr. cinsine aitdir. Penioillium funiculoamg Thom ile Aspergillus wentii Wehmer en yaygın türler olarak bulunmuştur. Tesbit edilen türlerin tümü Trakya Bölgesi için, Aspergillus pseudoglaucus Blochwitz türü, Aspergillus terricola var. inducus (M. and A.), n. Comb. Raper and Pennell ve Aspergillus terreus var. aureus Thom and Raper varyeteleri ile Penioillium montanense Christensen and Backus, Penioillium godlewskii Zaleski, Penioillium caaei Staub, Penioillium megasporum Orput and Pennell- ve Penioillium wortmanni Klöcker türleri Türkiye için yeni kayıtdır. Araştırma bölgesinde, 1 gr. toprakdaki mikrofungus sayısı 32.000 ile 864.OOO arasında değişmiş, genel ortalama ise 187.564,25 olarak tesbit edilmiştir. Mikrofunguslara en fazla sonbaharda, en az olarak da yazın rastlanmıştır. Toprak PH'sı ile sonbahar mikrofungus sayısı arasında ve toprakdaki fosfor miktarıyla ilkbahar mikrofungus sayısı arasında gerçek bir ilgi bulunmuştur. Toprağın organik maddesi, su ile doymuşluk oranı, pE'sı, toprakdaki fosfor ve potasyum değerlerinin her mevsim mikrofungus populasyonunâ ortak etkisi, sonbaharda % 35, kışın % 19, ilkbahar da % 14 ve yazın % 6 olarak tasbit edilmiştir.
