Ligamentum flavum hipertrofisinde potansiyel bir terapötik hedef olarak diskoidin domain reseptörü
Özet
Dejeneratif lomber dar kanal (DLDK) yaşlı hastalarda en sık karşılaşılan omurga
hastalığıdır. Kanal daralması, LF’un kalınlığının artmasına bağlıdır. Fibrozis ise LF’un
kalınlığının artmasının en önemli sebebidir. LF fibrozisinin altında yatan kesin moleküler
mekanizma tam olarak anlaşılamamıştır. Dejenerasyon altında, LF fibrozisinde elastin liflerin
azalması ve kollajen liflerin artması görülür. Mettaloproteiazlar (MMP) gibi kollajen reseptör
ailesi yeni kollajen sentezind hayati rolleri vardır. DDR1 ve DDR2 kollajen reseptör ailesinin
yeni tanımlanan üyeleridir.
Bu çalışmanın amacı, LF fibrozunun DDR1 ve DDR2 gen ekspresyon modellerini
değerlendirmektir. İnsan LF doku örnekleri, dejeneratif lomber dar kanal (DLDK) (n = 23)
veya lomber disk hernisi (LDH) (n = 12) nedeniyle opere edilen 35 hastadan elde edildi. LDH
hastaları kontrol grubu olarak alındı. Fibrozis derecesi Masson'un trikrom boyaması ile
gösterildi. DDR1 ve DDR2'nin proteinlerinin ekspresyon profili, polimeraz zincir reaksiyonu
(PCR) ve western blotlama ile analiz edildi. DLDK grubundaki tüm dokularda fibrosis
derecesi artmış olarak tespit edildi. DLDK grubunda DDR1 ve DDR2 gen ifadesi saptandı.
Benzer şekilde, western blod analizleri DLDK grubunda DDR1 ve DDR2 genlerinin
ifadelerinin yükseldiğini göstermiştir.
Sonuç olarak, çalışmamızın sonuçları DDR1 ve DDR2'nin LF fibrozusunun moleküler
mekanizması üzerinde önemli bir rolü olabileceğini ve DLDK tedavisi için ilaç geliştirme
stratejilerinde önemli bir hedef olabileceğini ortaya koymuştur. Degenerative lumbar canal stenosis (DLCS) is the most common spinal disorder in the
elderly patients. Canal narrowing resutls from increased thickness of ligamentum flavu (LF).
Fibrosis is the main cause of thickness of LF. The underlying exact molecular mechanism(s)
of LF fibrosis is not completely understood. Under the degeneration, the fibrotic change of LF
included decreased elastic fibers and increased collagen fibers. The collagen receptros family,
involved mettaloproteinase (MMP), are playing pivotal role in the synthesis of novel collagen
receptors. Both of DDR1 and DDR2 are a novel member of family of collagen receptors.
The aim of this study was to identify the DDR1 and DDR2 gene expression patterns of
the LF fibrosis. The human LF specimens were obtained from 35 patients who underwent
lumbar spine surgery for either degenerative lumbar canal stenosis (DLCS) (n=23) or lumbar
disc herniation (LDH) (n=12). In this design, patients with LDH served as the control group.
The degree of fibrosis was demonstrated by Masson’s trichrome staining. The location and
expression profiling of the DDR1 and DDR2 were analyzed by quantitative real-time
polymerase chain reaction and Western blotting. The grade of LF fibrosis was observed in all
samples of the DLCS group. DDR1 and DDR2 was detected in the DLCS group. Similarly,
western blod anaylses showed that the experion of DDR1 and DDR2 regulation in the DLCS
group.
Consequencly, the results of our study revealed that may DDR1 and DDR2 may be a
pivotal role on the molecular mechanism of the LF fibrosis, and be significant targets in drug
development strategies for the treatment of DLCS.
