Osteoporoz tayinine yönelik biyosensör sistemi geliştirilmesi
Özet
Osteoporoz, belirti vermeyen bir hastalık olup birçok kişi hastalığının farkında
değildir. Kemikte kırık meydana gelmeden önce sessiz dönemde osteoporoz tanısının
konulması son derece önemlidir. Kemik metabolizma ve döngüsünün
değerlendirilmesinde kemik döngüsündeki yapım ve yıkım ürünleri biyokimyasal
belirteç olarak kullanılmaktadır. Osteoporoz tanısı ve takibinde klinik muayene,
görüntüleme yöntemlerinin yanısıra bu biyokimyasal belirteçlerden yararlanılır.
Biyobelirteç temelli bu yöntemlerin çoğu özel ekipman ve uzman personel gerektiren,
zaman alıcı yöntemler olduğundan, alternatif olarak biyobelirteçlerin tayini için daha
duyarlı, daha kısa sürede uygulanabilen biyosensör sistemlerinin geliştirilmesi önem
kazanmaktadır.
Osteokalsin (OCN) ve Osteopontin (OPN) molekülleri bazı metabolik kemik
hastalıklarında anormal kemik kaybını öngörebilen biyolojik belirteçlerdendir. Bu tez
kapsamında; osteoporoz tanısı, seyri ve tedavi takibinde kullanılmak üzere immünolojik
temelli OCN ve OPN biyosensör sistemleri geliştirildi. Geliştirilen OCN ve OPN
biyosensörleri ile elektrokimyasal impedans spektroskopisi (EIS) kullanılarak 10-60
pg/µL tayin aralığında spesifik ve duyarlı OCN ve OPN ölçümleri yapıldı. Geliştirilen
biyosensörlerin tekrarlanabilirliklerinin yüksek olduğu belirlendi. Ayrıca bu
biyosensörler, yapay serum örneklerinin analizinde de başarıyla kullanıldı. Osteoporosis is a symptom-free disease and many people are not aware of the
disease. The diagnosis of osteoporosis in the silent period before the fracture occurs is
extremely important. The production and degradation products in the bone cycle are
important in the evaluation of bone metabolism and cycle as biochemical markers.
Today, in addition to the clinical examination and imaging methods, these biochemical
markers are used in the diagnosis and follow-up of osteoporosis. Since most of these
methods based on biomarkers are time-consuming methods that require special
equipment and expert personnel, the development of biosensor systems that are more
sensitive and fast becomes more important for the determination of biomarkers.
Osteocalcin (OCN) and Osteopontin (OPN) molecules are biomarkers that can
predict abnormal bone loss in some metabolic bone diseases. In this thesis, OCN and
OPN biosensor systems based on immunoassay were developed for the diagnosis,
prognosis and follow-up of osteoporosis. By using the developed OCN and OPN
biosensors, the OCN and OPN determinations were performed specifically and
sensitively using electrochemical impedance spectroscopy (EIS) within the
concentration range of 10-60 pg/µL. The reproducibilities of the developed biosensors
were determined to be high. Also, these biosensors were also used successfully in the
analysis of artificial serum samples.
